Intermediate filaments are a very broad class of fibrous proteins that play an important role as both structural and functional elements of the cytoskeleton. Ranging in size from 8 to 12 nanometers (in diameter; see Figure 1), intermediate filaments function as tension-bearing elements to help maintain cell shape and rigidity, and serve to anchor in place several organelles, including the nucleus and desmosomes.

They are rope-like structures formed by overlapping bundles of proteins. They are 10 nm in diameter and are more stable compared to actin filaments. Intermediate filaments function in the maintenance of cell-shape by bearing tension. Our interest in the structure and function of intermediate filament proteins was initiated approximately 7 years ago while working on the regulation of macromolecular synthesis in picornavirus-infected mam­ malian cells.

Think of them as the small pickup truck of these fibers. Intermediate filaments are about 10 nm in diameter with wavy profiles in electron micrographs of thin sections of cells (Fig. 35-1B) or after negative staining isolated filaments (Fig. 35-3B). In some cases, the N- or C-terminal domains project from the surface (Fig. 35-3C).

Intermediate filaments are the structures that form a network around the nucleus and extend to the periphery of the cell. Diameter of each filament is about 10 nm.

The apparent diameter was reported to be in the range of 10 nm and, therefore, “intermediate” between that of actin filaments (6 nm) and myosin filaments (15 nm). This criterion was then used to investigate other tissues, as well as cultured cells, for the presence of 10-nm-type filaments ( Goldman 1971 ; Small and Sobieszek 1977 ).

The subunits of intermediate filaments are elongated, not globular, and are associated in an antipolar manner. As a result, the overall filament has no polarity, and therefore no motor proteins move along intermediate filaments. Intermediate filaments are found only in complex multicellular organisms.
